Innovative strategies are emerging in the quest to conquer resistance mechanisms in cancer treatments. The TAM (TYRO3, AXL, and MERTK) family receptor tyrosine kinases (RTKs) have gained attention as potential therapeutic targets across various cancers. These RTKs trigger pathways that bolster cancer cell survival, metastasis, and resistance to therapies. Additionally, they play a role in suppressing antitumor immunity. TAM antagonists hold significant promise, offering both direct and immune-mediated therapeutic benefits, especially when combined with existing cancer treatments. This review equips clinicians with insights into TAM RTKs' potential and ongoing developments in targeting them for cancer therapy.
