The hope and hype of ellagic acid and urolithins as ligands of SARS-CoV-2 Nsp5 and inhibitors of viral replication

Non-structural protein 5 (Nsp5) is a crucial player in SARS-CoV-2 replication, aiding the virus in evading the immune system. Scientists are now exploring the use of natural compounds to inhibit Nsp5 and develop antiviral agents. A recent study delved into the properties of ellagic acid and its gut metabolites, urolithins A-D, as potential Nsp5 ligands. The results revealed urolithin D as a promising Nsp5 ligand, boasting potent binding capabilities in the nanomolar range. However, when tested in a Vero E6 assay against SARS-CoV-2, urolithin D showed limited viral replication inhibition. These findings shed light on the potential of natural products as viral protease inhibitors, while also emphasizing the need for further research to fully harness their antiviral properties.
