
Chemotherapy has been found to alter the tumour microenvironment in pancreatic ductal adenocarcinoma (PDAC) by reducing the expression of inhibitory checkpoint molecules, particularly TIGIT, on CD8+ T cells. This discovery suggests that blocking TIGIT before or at the same time as chemotherapy could be more effective than current immunotherapy approaches in PDAC. This conclusion was reached based on single-cell RNA sequencing of PDAC samples taken before and after chemotherapy.
