Tolebrutinib Shows Comparable Relapse Rates to Teriflunomide in MS Trials, May Slow Disability Progression

In two phase 3 trials (GEMINI 1 & 2), tolebrutinib, an oral BTK inhibitor, showed no superiority over teriflunomide in reducing relapse rates in relapsing multiple sclerosis. However, pooled data suggested tolebrutinib may lower sustained disability progression (HR: 0.71). Safety profiles were similar, with minor bleeding more frequent in the tolebrutinib group.
