
Immunotherapy holds great promise in combating human papillomavirus (HPV)-associated cancers, which create a suppressive tumor microenvironment. A study investigated the potential of using valproic acid (VA) as an epigenetic agent in combination with avelumab to boost anti-tumor immunity in HPV-associated solid tumors. The study analyzed RNA sequencing data from peripheral blood mononuclear cells (PBMCs) in non-responders (NRs) and responders (Rs) to treatment. Results revealed distinct immune response patterns, with a significant upregulation of NETosis-related transcripts in NRs. Elevated IL-8/IL-18 cytokines and transcriptome signatures also differentiated NRs from Rs. Plasma IL-8/IL-18 and PBMC RNA-seq might serve as predictive biomarkers for immunotherapy outcomes.
