Valproate Linked to Cortical Thinning in Juvenile Myoclonic Epilepsy

A study examined the impact of valproate on cortical morphology in individuals with juvenile myoclonic epilepsy (JME). Using T1-weighted brain imaging, cortical thickness was analyzed in 36 JME patients on valproate, 36 JME patients not on valproate, 36 controls, and 19 individuals with temporal lobe epilepsy (TLE) on valproate. The study found significant thinning of the precentral gyri in JME patients on valproate compared to controls and those not on valproate. Additionally, a dose-dependent negative correlation was observed between valproate use and cortical thickness in the precentral and postcentral gyri and superior frontal gyrus. This effect was not seen in TLE patients on valproate. The findings suggest valproate is a key modulator of cortical morphology in JME, potentially contributing to its therapeutic efficacy.
