Vorasidenib in IDH1- or IDH2-Mutant Low-Grade Glioma

Vorasidenib, an oral inhibitor of mutant IDH1 and IDH2 enzymes, demonstrated promising results in a double-blind, phase 3 trial involving patients with residual or recurrent grade 2 IDH-mutant glioma. Patients were randomly assigned to receive either vorasidenib or placebo, with imaging-based progression-free survival as the primary endpoint. The study found that vorasidenib significantly improved progression-free survival compared to placebo (median progression-free survival of 27.7 months vs. 11.1 months) and delayed the time to the next anticancer intervention. Adverse events were reported, with a higher occurrence in the vorasidenib group, including increased alanine aminotransferase levels. These findings suggest that vorasidenib holds promise as a treatment option for IDH-mutant gliomas.
