Whole-Genome Methylation Sequencing Uncovers Persistent Epigenetic Dysregulation in COVID-19 Patients

Researchers have found that patients with severe COVID-19 infections exhibit abnormal DNA methylation long after recovering from the disease. Using DNA methylation arrays and next-generation sequencing technologies, the study compared paired longitudinal samples from COVID-19 patients with their methylome obtained one year after hospital discharge and with methylome profiles from healthy individuals. The study found that differentially methylated regions (DMRs) that did not return to baseline after one year were associated with persistently dysregulated immune and inflammatory profiles, while those that returned to baseline were associated with viral responses, including interferon signaling. These findings suggest that epigenetic mechanisms, such as DNA methylation, may play a role in perpetuating long-term disordered inflammatory responses in COVID-19 patients.
