Aging Gracefully? Not for Your Gums: Cellular Senescence in Periodontitis

The study evaluates how accelerated cellular senescence, a manifestation of aging at the cellular level, maybe a crucial process in the progression of periodontitis due to the damaging effects of risk factors like a persistent bacterial infection, chronic inflammation, diabetes, and smoking. The accumulation of senescent cells leads to tissue destruction, limits regeneration, exacerbates inflammation, remodels tissues, and is a source of chronic inflammation. The study proposes the acronym ALTER to summarize key senescence-associated features implicated in tissue deterioration. The review suggests that the accelerated activation of cellular senescence could be a central underlying mechanism for periodontitis progression, disrupting local homeostasis, and contributing to periodontal destruction. Understanding the host contribution to periodontal destruction through premature DNA damage-driven cellular senescence will help develop new therapeutic strategies.
