Alveolar macrophages drive lung fibroblast function in cocultures of IPF and normal patient samples

The article discusses a study on the cellular interactions between alveolar macrophages (AM) and fibroblasts in idiopathic pulmonary fibrosis (IPF). The researchers isolated cells from IPF and normal human lung tissue and studied their interactions in various culture conditions. They found that normal AMs downregulated extracellular matrix gene expression, reduced baseline contractility of normal fibroblasts, and had little effect on IPF fibroblasts. IPF AMs had little to no effect on ECM gene expression or contractility of either normal or IPF fibroblasts. Stimulation with Toll-like receptor (TLR) agonists increased the production of pro-inflammatory cytokines TNFα and IL-1β in IPF AMs compared to normal AMs, and fibroblast behavior was influenced by cell proximity. These findings suggest that normal fibroblasts are more responsive to AM crosstalk, and AM influence on fibroblast behavior depends on cell proximity.
