
In a groundbreaking study, researchers have discovered the potential of oral tofacitinib as an effective treatment for multiple keloids, a challenging clinical issue. Seven patients were given 5 mg of tofacitinib orally twice a day for 12 weeks. Remarkable improvements were noted in the Patient and Observer Scar Assessment Scale (POSAS), Vancouver Scar Scale (VSS), and Dermatology Life Quality Index (DLQI) scores, indicating reduced keloid volume, height, and dermis thickness. Immunohistochemistry revealed decreased collagen synthesis and fibroblast proliferation. Tofacitinib inhibits these processes via suppression of STAT3 and SMAD2 pathways, offering hope for painless and convenient keloid treatment and enhancing patients' quality of life.
