This study investigated the potential of nasal administration of live attenuated Streptococcus pneumoniae (Spn) strains in preventing pneumococcal pneumonia. Healthy adults were randomized for nasal administration with wild-type Spn or genetically modified Spn strains. Results demonstrated the safety of nasal colonization with live attenuated Spn and significant protection against recolonization with wild-type Spn. The findings suggest a promising strategy for preventing pneumococcal infections through nasal administration of genetically modified live attenuated Spn strains, emphasizing their potential in enhancing protective immunity.
