Romosozumab treatment aids hip fracture prevention by boosting bone density and formation through sclerostin protein inhibition. Study investigates human femur bone formation patterns and osteocyte expression, linking new bone buds to higher mechanical stress areas and low sclerostin expression. Patients with hip fractures exhibit active modeling-based formation in femoral trabeculae, particularly in high-stress zones. Greater bud density corresponds to improved trabecular metrics (BV/TV, BS/TV, Tb.Th, Tb.N, Conn.D), and reduced trabecular separation (Tb.Sp). Findings suggest modeling-based bone formation as a therapeutic target for enhancing bone structure in hip fracture patients.
