CAR-T cells dual-target CD123 and NKG2DLs to eradicate AML cells and selectively target immunosuppressive cells

The study has developed 123NL CAR-T cells targeting CD123 and NKG2DL in acute myeloid leukemia (AML). Overcoming AML's immunosuppressive environment, these cells eradicate AML cells while selectively attacking immunosuppressive M-MDSCs and M2 cells. Incorporating a compact marker/suicide gene enhances precision. The binding of Rituximab to the gene halts CAR-T cells' activity. This breakthrough offers a dual strategy to combat AML and evade target escape in a hostile microenvironment.
