Cell‐type specific determination of reactive oxygen species by flow cytometry

The study aimed to determine fluorescence intensity cut-off values for reactive oxygen species (ROS) in seminal cells, which could differentiate between samples with ROS overproduction caused by leukocytes and normal samples. Semen samples were obtained from patients undergoing andrology consultations, and routine analyses were conducted following World Health Organization guidelines. The results showed that leukocytospermic samples had higher ROS-related fluorescence intensity in both sperm and leukocytes compared to normozoospermic samples. There was a positive and linear correlation between fluorescence intensity in sperm and leukocytes. The findings suggest that leukocytes may contribute significantly to the overall ROS levels measured in sperm, impacting oxidative stress. Reliable differentiation between ROS-overproducing leukocytospermic and normozoospermic samples is possible based on ROS fluorescence intensity measurements.
