CRISPR/Cas9 screen for genome‐wide interrogation of essential MYC‐bound E‐boxes in cancer cells

The transcription factor MYC is a key player in cancer development, binding to specific DNA sequences (E-boxes) to regulate gene expression in various cancers. Researchers used CRISPR/Cas9 to disrupt E-boxes and the Brunello library to knockout genes, screening MYC-dependent cancer cell lines (K562, ST486, HepG2, MCF7). They identified essential E-boxes and genes, common and cell-specific, crucial for cancer pathways. Validation confirmed the disruption's impact on MYC binding, gene expression, cell growth in vitro, and tumor growth in vivo. This validated approach offers new insights into MYC-related cancer vulnerabilities, presenting a promising tool for studying such vulnerabilities in cancer cells.
