Detecting ALK, ROS1, and RET fusions and the METΔex14 splicing variant in liquid biopsies of non‐small‐cell lung cancer patients using RNA‐based techniques

In this study, researchers investigated the effectiveness of different technologies for detecting gene fusions and splicing variants in liquid biopsies of cancer patients. Next-generation sequencing (NGS) is commonly used but lacks sensitivity in liquid biopsies. The study found that nCounter, a molecular profiling platform, successfully detected these alterations in liquid biopsies and correlated well with tissue genotyping. Additionally, digital PCR (dPCR) was found to be useful for monitoring disease progression in patients with fusion-specific alterations. The study specifically focused on ALK, ROS1, RET, and MET∆ex14 gene fusions, which are associated with targeted therapy response in non-small-cell lung cancer. The researchers concluded that nCounter can be employed for multiplex detection of fusion and MET∆ex14 transcripts in liquid biopsies, while dPCR can be utilized for disease follow-up. Circulating-free RNA (cfRNA) was recommended over extracellular vesicle RNA (EV-RNA) for these analyses.
