DNA Methylation Disrupted by Sleep Loss

DNA methylation analysis revealed that 14% of sites exhibited diurnal rhythms under a normal 24-hour sleep/wake cycle, which were significantly disrupted during 29 hours of sleep deprivation. As sleep deprivation progressed, hypomethylation dominated, with enrichment of genes linked to the FAS immune response pathway. This suggests that prolonged wakefulness alters DNA methylation patterns, potentially mediating immune response changes. The findings underscore the need to consider diurnal variation when using DNA methylation as a biomarker.
