DOCK2 Promotes Asthma Development by Eliciting Airway Epithelial–Mesenchymal Transition

A study indicated Epithelial–mesenchymal transition (EMT) is a pivotal contributor to the airway remodeling seen in asthma. The analysis discovered that DOCK2 (dedicator of cytokinesis 2), known for its role in vascular remodeling, is significantly induced in normal human bronchial epithelial cells exposed to house dust mite (HDM) extract and human asthmatic airway epithelium. Moreover, DOCK2 responds to TGF-β1, playing a pivotal role in EMT. Knocking down DOCK2 inhibits EMT, while its overexpression promotes it. DOCK2 deficiency in HDM-induced asthma models reduces EMT, and subepithelial fibrosis, and enhances pulmonary function. This study identifies DOCK2 as a potential therapeutic target for asthma treatment.
