Dose Reductions Do Not Affect Adjuvant Abemaciclib's Efficacy in High-Risk Early Breast Cancer

In the MonarchE trial, adjuvant abemaciclib demonstrated significant improvements in invasive disease-free survival (IDFS) and distant relapse-free survival (DRFS), extending beyond the 2-year treatment period for patients with high-risk early breast cancer. Exploratory analyses assessed the impact of dose reductions on efficacy across three patient subgroups based on relative dose intensity. The estimated 4-year IDFS rates remained consistent across all subgroups. Time-dependent Cox proportional hazard models indicated that the efficacy of abemaciclib was not compromised by dose reductions. These findings suggest that protocol-mandated dose reductions did not affect the efficacy of adjuvant abemaciclib in patients with high-risk early breast cancer characterized by node-positive, hormone receptor-positive, human epidermal growth factor 2-negative tumours.
