
This study aimed to assess the reliability of cancer-associated fibroblast (CAF)-HSVtk suicide system in a co-culture model of oral cancer cells. CAFs were modified with PCDH-HSVtk and treated with ganciclovir (GCV) to selectively eliminate CAFs. The expression of HSVtk in CAFs was significantly higher compared to the control group, and the survival rates of CAFs with GCV were significantly reduced. After selectively depleting CAFs, the growth and migration rates of co-cultured oral cancer cells were decreased. However, oral tumor cell death was not affected. These findings suggest that the CAFs-HSVtk system can be a useful model for studying CAF characteristics in oral cancer.
