
Recent randomized controlled trials (RCTs) have examined the effectiveness and safety of newly developed once-weekly basal insulin analogues in managing glycemic control. A systematic review of nine RCTs involving 3,962 patients with type 2 diabetes found that these once-weekly insulins outperformed once-daily analogues in reducing glycated hemoglobin (HbA1c) levels and increasing time in the target range. Notably, icodec demonstrated the most significant HbA1c reduction and increased odds of achieving HbA1c <6.5%. While once-weekly insulins were associated with more level 1 hypoglycemia events, they proved safer for level 2 or 3 nocturnal hypoglycemia.
