
A transcriptomic analysis revealed FAM114A1 as a key protein associated with membranous nephropathy (MN), with its expression significantly upregulated in both human MN and rat passive Heymann nephritis (PHN). Immunofluorescence staining showed that FAM114A1 localized primarily to podocytes' primary and foot processes. In silico modeling revealed FAM114A1 as an all-alpha protein with several conserved regions. Silencing FAM114A1 in cultured podocytes disrupted cytoskeletal development, cell migration, and attachment. Affinity pulldown assays identified FAM114A1’s interaction with cytoskeleton-associated proteins, suggesting its involvement in podocyte integrity and glomerular injury response.
