
The study investigated the role of general control nonderepressible 2 (GCN2) in regulating diurnal patterns of hepatic integrated stress response (ISR) and metabolism. Researchers hypothesized that GCN2 activation cues hepatic ISR signaling over a 24-hour feeding-fasting cycle. Using wild-type (WT) and GCN2 knockout (KO) mice, they examined the effects of a leucine-devoid diet (LeuD). Results showed that GCN2 activation maintained diurnal ISR in WT mice, while GCN2 KO mice displayed disrupted metabolic rhythms, indicating GCN2's essential role in nutrient stress and metabolic regulation.
