Glioblastoma (GBM) is a highly aggressive and treatment-resistant cancer due to its complex heterogeneity at the cellular level. The tumor's diverse cell states, composition, and phenotypical characteristics make classification and effective therapies challenging. Recent advancements in sequencing technologies have revealed the single-cell heterogeneity of GBM, shedding light on its resistance mechanisms and potential therapeutic targets. Moreover, heterogeneity varies between new and recurrent GBM, as well as treatment-naïve and experienced patients. Understanding the intricate cellular network underlying GBM heterogeneity is crucial for developing new strategies to combat this lethal disease. This overview discusses the multifaceted layers of GBM heterogeneity and highlights recent discoveries enabled by single-cell technologies.
