
A study investigated the role of the Carbohydrate Response Element (ChoRE) Binding Protein (ChREBP) and Max-like protein X (MLX) in glucose-induced lipogenic gene transcription, particularly in Hepatocellular Carcinomas (HCCs). Elevated MLX expression in HCC specimens is observed, and downregulating MLX inhibits HCC cell proliferation. Liver-specific Mlx knockout in mice significantly reduces lipogenic gene expression and circulating lipid levels, effectively blocking HCC tumor development in multiple models. A high-fat diet partially restores tumorigenesis in Mlx-deficient livers. These findings suggest ChREBP: MLX as potential targets for HCC cancer therapies.
