Impact of rituximab plasma concentration on major relapse risk in ANCA-associated vasculitides during maintenance therapy

The study explored the relationship between rituximab plasma concentration, genetic polymorphisms in PK/PD candidate genes, and clinical outcomes in patients with antineutrophil cytoplasmic antibody (ANCA)‐associated vasculitides. Patients were randomized to receive a fixed‐schedule RTX infusion or an individually‐tailored regimen. Results showed that low RTX plasma concentration at 3 months was an independent risk factor for major relapse. Drug monitoring could be useful to individualize the schedule of rituximab administration within the maintenance phase. STAT4 rs2278940 and PRKCA rs8076312 were significantly associated with rituximab plasma concentration but not with major relapse onset.
