Inhibition of SUMOylation enhances DNA hypomethylating drug efficacy to reduce outgrowth of hematopoietic malignancies

This study investigates the combination of two drugs, TAK981 and 5-Aza-2’-deoxycytidine, for the treatment of MYC-driven hematopoietic malignancies, including Burkitt lymphoma and diffuse large B-cell lymphoma. The study found that the combination of the two drugs led to increased DNA damage and reduced cell proliferation in vitro. In vivo experiments showed a significant reduction in the outgrowth of Burkitt lymphoma in an orthotopic xenograft model. The study highlights the potential of combining drugs with complementary mechanisms to improve cancer treatment efficacy.
