JAK Inhibitors Do Not Increase GI Perforation Risk in RA

The systematic review and network meta-analysis of 23 RCTs (n=20,023) in RA patients found a GIP incidence of 0.19% with JAKi use. No significant risk increase was found compared to csDMARDs (RR = 1.02; 95% CI: 0.41–2.56). SUCRA rankings confirmed low GIP risk for JAKi (50.8%) and bDMARDs (77.0%). Findings support continued safety in clinical use.
