
A study highlights KLHL29 as a key tumor suppressor in Triple-Negative Breast Cancer (TNBC), regulating chemosensitivity. Lower KLHL29 levels were associated with unfavorable prognoses. Manipulating KLHL29 affected TNBC growth, migration, and invasion. KLHL29's role in degrading DDX3X destabilizes CCND1 mRNA, inducing cell cycle arrest. Combined with platinum-based chemotherapy, DDX3X inhibition using the RK33 compound demonstrated significant suppression of TNBC, presenting a potential breakthrough in treating the often challenging TNBC subtype.
