Long-Term Survival Benefits of Tremelimumab Plus Durvalumab and Chemotherapy in mNSCLC

In a study, tremelimumab plus durvalumab and chemotherapy (T+D+CT) significantly improved overall survival (OS) compared to chemotherapy alone (CT) in patients with EGFR/ALK-wild-type metastatic non-small cell lung cancer (mNSCLC) after a median follow-up of 63.4 months. The hazard ratio (HR) for OS with T+D+CT versus CT was 0.76 (95% CI: 0.64–0.89), with a 5-year OS rate of 15.7% versus 6.8%. Tremelimumab plus durvalumab and chemotherapy showed sustained benefits across various patient subgroups, including those with PD-L1 TC <1% and specific genetic mutations. No new safety concerns were identified, supporting the long-term use of T+D+CT as a first-line treatment for mNSCLC.
