
Microfracture, a surgical approach for osteochondral lesions of the talus (OLTs), often yields fibrocartilage with suboptimal mechanical properties. This study evaluated whether adding losartan, fisetin, or both could enhance cartilage repair post-microfracture in a rabbit model. Female rabbits with induced talar defects received daily oral doses of losartan (10 mg/kg), fisetin (20 mg/kg), or both for 12 weeks. Losartan and fisetin groups demonstrated improved cartilage repair, higher macroscopic and histological scores, and organized chondrocyte formation compared with microfracture alone. However, the combined treatment group (losartan+fisetin) showed no added benefits and lower histology scores. These findings suggest that losartan or fisetin individually may enhance microfracture-mediated cartilage repair in OLTs, while combined use lacks synergy.
