This study investigated the clinical and immunological implications of human leukocyte antigen (HLA) loss of heterozygosity (LOH) in head and neck squamous cell carcinoma (HNSCC). By analyzing genomic and transcriptomic data from 522 HNSCC tumors, the researchers found that HLA LOH was a common event occurring in 53% of tumors. Advanced-stage tumors had a higher frequency of HLA LOH. Tumors with HLA LOH exhibited reduced lymphocyte and T cell infiltration, indicating an immunologically suppressed tumor microenvironment. These findings suggest that HLA LOH plays a role in immune evasion and could serve as a predictive biomarker or therapeutic target in HNSCC.
