
The study determined the impact of insulin glargine U-100 and its metabolites on insulin sensitivity and β-cell measures in individuals with type 2 diabetes. The study used liquid chromatography-mass spectrometry to measure concentrations of endogenous insulin, glargine, and its two metabolites in plasma. It found incomplete cross-reactivity of glargine and its metabolites in the insulin immunoassay. This resulted in a bias in fasting-based measures of insulin sensitivity and β-cell function but not in dynamic insulin responses. The study suggests caution in interpreting fasting-based measures in individuals using insulin glargine U-100.
