Methionine's Critical Role in Cancer and Potential Therapy

Methionine has been identified as a key regulator of sulfur metabolism and tumor growth. Its role in the one-carbon metabolism cycle, alongside folate, influences S-adenosylmethionine (SAM) synthesis, essential for tumor progression. Methionine also regulates chromatin states depending on p53 status and facilitates m6A methylation of NR4A2 mRNA, promoting proliferation in esophageal carcinoma. In this context, Celecoxib, an NR4A2 inhibitor, shows promise as a potential cancer therapy. Furthermore, formaldehyde, both from endogenous and exogenous sources, directly affects SAM levels and one-carbon metabolism, with significant implications for cancer development. These findings offer novel insights into methionine's role in cancer and its therapeutic possibilities.
