Phase 1 identified a safe dose, 1.0 mg of Mezigdomide, to be taken once daily with dexamethasone in a 28-day cycle. In phase 2, 101 patients with triple-class-refractory myeloma participated, showing an overall response rate of 41%, with a median duration of response at 7.6 months. While common adverse effects included reversible neutropenia and infections, no unexpected toxicities emerged. This all-oral therapy offers hope for patients with limited treatment options for this incurable disease.
