Next-Gen KRAS-G12C Inhibitor Shows Promising Clinical Activity

D3S-001 delivers high response rates across KRASG12C cancers. D3S-001 (elisrasib), a next-gen KRAS-G12C inhibitor, showed strong efficacy and tolerability in a phase Ia trial of 42 patients with advanced NSCLC, CRC, or PDAC. Among 34 KRAS-G12C inhibitor-naive patients, the ORR reached 73.5%, with a DCR of 97.1%. No grade 4–5 TRAEs were reported. The data suggest D3S-001 may outperform first-generation inhibitors due to improved pharmacologic properties.
