In 2022, the European LeukemiaNet (ELN) unveiled its second update to the risk classification for Acute Myeloid Leukemia (AML), marking a significant milestone since its inception in 2010. ELN2022 designates "secondary-type" mutations (STMs) as adverse risk factors. This classification aligns with recent World Health Organization (WHO) and International Consensus Classification (ICC) updates. However, questions emerged about the interplay between STMs and favorable markers like NPM1 mutations. Research on a multicenter cohort of 936 NPM1-mutated AML patients revealed that STMs do not negate the favorable prognosis associated with NPM1 mutations. This study supports ELN's classification of NPM1-mutated patients as favorable, even in the presence of STMs.

A newly developed evidence-based guideline provides recommendations for diagnosing, staging, and managing keratoconus in children and adults. Using systematic evidence synthesis and the GRADE approach, it addresses corneal cross-linking, intrastromal corneal ring segments, keratoplasty, and individualized follow-up. Most recommendations remain conditional because evidence certainty is low or very low in several clinical areas.

UROLOGY

A nationwide Colombian study identified 4,608 healthcare encounters related to childhood interstitial and diffuse lung disease (chILD) from 2016–2024. Average annual prevalence was 3.32 per 100,000 children, highest among infants and with a secondary rise during adolescence. Reported cases increased annually (IRR 1.022), while declining markedly during the COVID-19 pandemic.
