Higher pancreatic fat drives beta cell dedifferentiation in diabetes. This study (n=50) found pancreatic fat was significantly higher in type 2 diabetes (10.24% vs. 0.74%, p<0.001) and correlated with beta cell loss (r=-0.675, p=0.006). Single-cell RNA-seq revealed increased ALDH1A3 expression, beta cell transdifferentiation, and immune activation. Adipocytes influenced immune recruitment, suggesting pancreatic fat may contribute to beta cell failure in type 2 diabetes.
