The use of genome-wide CRISPR–Cas9 knockout screens in BRCA1/2-proficient prostate cancer cells enabled the identification of previously unknown genes, the loss of which affects PARP inhibitor response. For example, MMS22L deletion caused PARP inhibitor hypersensitivity and CHEK2 conferred resistance.
