PCSK9 inhibition with orally administered NNC0385-0434 in hypercholesterolaemia

In a groundbreaking phase 2 study, the oral PCSK9 inhibitor NNC0385-0434 demonstrated significant efficacy in reducing LDL cholesterol over 12 weeks. The study included 267 patients across three dose cohorts, with reductions of 32.0%, 44.9%, and 61.8% compared to placebo. Notably, the 100 mg cohort showed LDL reductions similar to injectable evolocumab. Adverse events were mainly related to COVID-19 (12% incidence), with gastrointestinal disorders reported in both NNC0385-0434 and evolocumab groups. The findings suggest the oral inhibitor's promising efficacy and patient tolerance, mirroring injectable counterparts, marking a potential paradigm shift in PCSK9 inhibition
