
The phase 3 KEYNOTE-811 study found that pembrolizumab, combined with trastuzumab and chemotherapy, significantly improved progression-free survival in patients with metastatic HER2-positive gastro-oesophageal cancer. The study, conducted across 168 medical centers in 20 countries, involved 698 patients. In the second interim analysis, median progression-free survival was 10.0 months in the pembrolizumab group, compared to 8.1 months in the placebo group. Although overall survival showed improvement in the pembrolizumab group, it did not reach statistical significance. Grade 3 or worse treatment-related adverse events occurred in 58% of patients in the pembrolizumab group and 51% in the placebo group. The findings suggest a promising avenue for treatment but require further analysis for complete validation.
