Peritoneal high-fat environment promotes peritoneal metastasis of gastric cancer cells

Peritoneal metastasis (PM) is a serious form of gastric cancer (GC) that is difficult to treat and has a poor prognosis. The molecular mechanism underlying PM is not well understood. The study found that the RNA modification m5C, which is involved in tumor progression, plays a role in PM. Specifically, NSUN2, which is upregulated in PM, regulates ORAI2 mRNA stability through m5C modification, leading to increased ORAI2 expression and further promoting PM and colonization of GC. YBX1 binds to the ORAI2 m5C modification site and acts as a "reader." Additionally, the study found that peritoneal adipocytes provide fatty acids for GC cells, leading to the upregulation of E2F1 and NSUN2 through the AMPK pathway and the promotion of PM and colonization of GC.
