
Necrotizing granulomatous inflammation is characteristic of granulomatosis with polyangiitis (GPA), a form of anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis. However, the mechanisms underlying the development of granulomas are poorly understood. New research suggests that persistent antigenic stimulation by proteinase 3 (PR3) is the trigger for the formation of multinucleated giant cells (MGCs) and granuloma-like structures in GPA.
