Proteomic Insights into IL-6 Signaling in Atherosclerosis: Identifying CXCL10 as a Potential Therapeutic Target

Genetic and experimental evidence indicates the role of IL-6 signalling in atherosclerosis. While ongoing trials target IL-6, potential immune response issues need consideration. The study used genetic data from 522,681 individuals to create an IL-6R inhibition proxy. Mendelian randomization revealed effects on 70 proteins related to cytokine regulation and immune cell function. CXCL10 emerged as a mediator, with its decreased levels linked to reduced risk of cardiovascular diseases. Higher CXCL10 correlated with increased cardiovascular events and immune system activation in atherosclerotic lesions, suggesting it is a potential atheroprotective drug target.
