
Asthma, often caused by allergic inflammation, involves airway narrowing due to inflammation-induced smooth muscle contraction. While asthma treatment focuses on blocking inflammation, research into inflammation's effect on airway smooth muscle is limited. Gebski et al. investigated rhinovirus (RV) infection's impact on bronchodilation mechanisms. Using human precision-cut lung slices, airway epithelial cells, and smooth muscle cells, they found RV-C15 infection reduced bronchodilation responses. This was attributed to an epithelial cell-derived factor impairing G protein-mediated coupling and increasing GNAI1 gene expression, hindering bronchodilation pathways. This work sheds light on virus-induced asthma exacerbation mechanisms and could inspire new treatments. However, challenges like immune system absence in models remain to be addressed.
