A groundbreaking study explored the use of RNA-engineered chimeric antigen receptor (CAR) T cells to treat autoimmune diseases safely. The study focused on individuals with myasthenia gravis, a disease mediated by pathogenic plasma cells. Descartes-08, an autologous anti-BCMA rCAR-T therapy, was administered to participants in a phase 1b/2a trial. No dose-limiting toxicity or neurotoxicity was observed, and adverse events such as headache, nausea, and fever were transient. Encouragingly, participants experienced significant improvements in myasthenia gravis severity scales after receiving Descartes-08 infusions for up to 9 months. The study suggests rCAR-T therapy could be a promising treatment option for autoimmune diseases, warranting further investigation.
