In phase 4 clinical trial, researchers examined the safety and effectiveness of a heterologous booster of the Ad5-nCoV vaccine following three doses of inactivated SARS-CoV-2 vaccine. Participants aged 18-80 were divided into two groups, receiving either Ad5-nCoV or inactivated vaccine. Predictable and manageable adverse reactions were observed. Ad5-nCoV induced significantly higher IgG levels and pseudovirus-neutralizing antibodies than the inactivated vaccine, maintaining high antibody levels for 90 days. Seroconversion after Ad5-nCoV booster was 71.4%, while inactivated vaccine had 5.2%. This suggests that Ad5-nCoV is more immunogenic and efficient as a fourth dose than inactivated vaccines, offering stronger and longer-lasting protection.
