Sodium Oxybate Effective for Alcohol-Responsive Laryngeal Dystonia

A phase IIb randomized, double-blind, placebo-controlled trial evaluated sodium oxybate for treating isolated laryngeal dystonia (LD). Conducted at Massachusetts Eye and Ear (2018–2021), the study involved 106 patients, categorized as alcohol-responsive (EtOH+) or alcohol-non-responsive (EtOH−). Sodium oxybate significantly improved symptoms in EtOH+ patients compared to placebo, with a 16% minimum efficacy threshold (OR = 2.09; p = 0.036) and an average symptom reduction of 40.81%. Effects peaked at 40 minutes post-dose and declined by 300 minutes without rebound symptoms. Safety assessments showed no significant cognitive or vital sign changes, though mild dizziness, nausea, and daytime sleepiness were reported. Sodium oxybate may be a promising therapy for EtOH+ LD patients.
