This study investigates plasma microRNA (miRNA) changes and metabolite levels following anterior cruciate ligament (ACL) reconstruction surgery. Examining 43 patients, miRNA sequencing and metabolomics identified significant alterations at 2 weeks post-surgery, with 46 miRNAs and 13 metabolites showing notable changes. Females exhibited higher levels of specific miRNAs and lower levels of certain metabolites than males post-surgery. Computational analyses revealed potential pathways influencing these changes, impacting post-traumatic osteoarthritis (PTOA) development. Understanding these early molecular responses could refine treatment strategies and mitigate PTOA risk after ACL reconstruction.
